ABSTRACT

Objective:To investigate the effect and mechanism of Chaihu Jia Longgu Mulitang (CJLM) on hippocampal NOD-like receptor protein 3 (NLRP3)inflammasome pathway in rats with depression. Method:Sixty male SD rats were randomly divided into a normal group,a model group, a MCC950 (1 mg·kg<sup>-1</sup>) group, and high- (13 g·kg<sup>-1</sup>), medium- (6.5 g·kg<sup>-1</sup>), and low-dose (3.25 g·kg<sup>-1</sup>) Chaihu Jia Longgu Mulitang groups, with 10 rats in each group.The depression model was induced by isolation combined with chronic unpredictable mild stimulation(CUMS) in rats except for those in the normal group. Rats were treated correspondingly for 21 days by intraperitoneal injection in the MCC950 group and gavage in other groups. The normal group and the model group received an equal volume of normal saline. The depression-like behaviors of rats were observed by sucrose preference test (SPT) and novelty-suppressed feeding test. Enzyme-linked immunosorbent assay (ELISA) was used to determine the levels of interleukin-1<italic>β </italic>(IL-1<italic>β</italic>) and IL-18 in the hippocampus of depressed rats. Western blot was used to detect the protein levels of NLRP3, apoptosis-associated speck-like protein containing a C-terminal caspase recruitment domain (ASC), and Caspase-1. Result:Compared with the normal group, the model group showed decreased sucrose preference rate (<italic>P</italic><0.01), prolonged novelty-suppressed feeding time (<italic>P</italic><0.01), enhanced protein expression of NLRP3,ASC, and caspase-1<italic> </italic>(<italic>P</italic><0.05, <italic>P</italic><0.01), and elevated expression of IL-1<italic>β</italic> and IL-18 (<italic>P</italic><0.01). Conclusion:CJLM can alleviate depression-like behaviors in CUMS-induced model rats, and the underlying mechanism is related to the inhibition of the NLRP3 inflammasome pathway.

ABSTRACT

Objective:To study the effect of modified Xiao Chaihutang on the expressions of excitatory amino acid transporters(EAATs) and vesicle glutamate transporters(VGLUTs)in hippocampus of rats with chronic depression, in order to explore the anti-depressant mechanism of modified Xiao Chaihutang based on glutamate transport. Method:A total of 120 SD rats were randomly divided into normal group, model group, and low, middle and high-dose modified Xiaochaihutang groups (6.5, 13, 26 g·kg-1) and riluzole group, with 20 rats in each group.Except normal group, the depression model of rats was prepared through Chronic restraint stress(CRS). The normal group and the model group were intragastrically (ig) given normal saline. The modified Xiao Chaihutang groups were intragastrically given corresponding herbal drugs (6.5, 13, 26 g·kg-1), and the Riluzole group was given Riluzole 20 mg·kg-1 through intraoeritoneal injection for 21 days, once a day. Then the depressive behaviors of rats were observed by forced swimming test (FST) and tail suspension test (TST). The level of glutamic acid (Glu) in rats hippocampus was determined by high performance liquid chromatography (HPLC). The mRNA expressions of EAAT1, EAAT2 and EAAT3 in hippocampus were detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR)method. Western blot was used to detect the protein expressions of EAAT1, EAAT2, EAAT3, VGLUT1 and VGLUT2 in rat hippocampus tissue. Nissl staining was used to observe the morphology of hippocampal neurons in rats. Immunohistochemical(IHC)S-P method were used to detect the location expressions of EAAT1, EAAT2 and NeuN proteins in rat hippocampal CA1 region tissue. Result:The immobility times in FST and TST were increased significantly(P<0.01), the mRNA and protein expressions of EAAT1,EAAT2,EAAT3 were decreased significantly (P<0.01), and as well as the expressions of VGLUT1 and NeuN were decreased significantly(P<0.01), while the level of Glutamate and the expression of VGLUT2 were increased significantly(P<0.01) in model group, compared with normal group. Compared with model group,the immobility times in FST and TST were decreased significantly(P<0.05, P<0.01), mRNA and protein expressions of EAAT1,EAAT2,EAAT3 were increased significantly(P<0.01), and expressions of VGLUT1 and NeuN were increased significantly(P<0.01). However, the level of Glutamate and the expression of VGLUT2 were decreased significantly(P<0.01), and the damage of hippocampal neurons in rats was mild in middle and high-dose modified Xiao Chaihutang groups. Conclusion:Modified Xiao Chaihutang has an anti-depressive effect. Its mechanism may be related to its up-regulation of expressions of EAAT1, EAAT2, EAAT3 genes and VGLUT1 protein in the hippocampus of depression model rats.

ABSTRACT

Objective:To study the effect of modified Suanzaoren Tang on the expression of excitatory amino acids receptor(EAARs) in hippocampus of rats with chronic depression, and to explore the anti-depressant mechanism of modified Suanzaoren Tang based on excitatory amino acids receptor. Method:Sixty SD rats were randomly divided into normal group,model group,and low,middle and high-dose modified Suanzaoren Tang groups,and ketamine group,with 10 rats in each group.Except normal group,the depression model of rats was prepared by using chronic restraint stress(CRS).The normal group and model group were intragastrically(ig) given normal saline.the modified Suanzaoren Tang groups were intragastrically given corresponding herbal drugs 6,12,24 g·kg-1, ketamine group group were given ketamine 0.015 g·kg-1 through intraoeritoneal injection,for 21 days,once a day.Then the depressive behaviors of rats were observed by Morris water maze and novelty feeding experiment.Western blot was used to detect the levels of DAR1,NMDAR2A,NMDAR2B,GluR1,mGluR1,CaMKⅡα and CaMKⅡβ protein expression in rat hippocampus tissue. Result:Compared with normal group,the time of novel ingestion and escape latencywere prolonged significantly(P<0.01), and the time of space exploration was shortened significantly(P<0.01).The levels of NMDAR1,NMDAR2A,NMDAR2B,mGluR1 and CaMKⅡβ expression were increased significantly(P<0.01),while the levels of GluR1 and CaMKⅡα expression were decreased significantly(P<0.01)in model group. Compared with model group,the time of novel ingestion and escape latency were shortened significantly (P<0.01), and the time of space exploration was prolonged significantly(P<0.01).The levels of NMDAR1,NMDAR2A,NMDAR2B,mGluR1 and CaMKⅡβ protein expression were decreased significantly(P<0.01),but the levels of GluR1 and CaMKⅡα expression were increased decreased significantly(P<0.01)in middle and high-dose modified Suanzaoren Tang groups. Conclusion:Modified Suanzaoren Tang can improve the behavior of chronic depression rats effectly. Its mechanism may be related with reduction the expression of NMDAR1,NMDAR2A,NMDAR2B,mGluR1 and CaMKⅡβ protein ,increase the expression of GluR1and CaMKⅡα protein.

ABSTRACT

Objective:To study the effect of Chaihu Jia Longgu Mulitang on phosphatidylinositol-3-kinases (PI3K)/protein kinase B (Akt)/glycogen synthase kinase 3β (GSK3β)/β-catenin signaling pathway of hippocampus in rats with depression. Method:A total of 120 SD rats were randomly divided into normal group,model group, and low, middle and high-dose Chaihu Jia Longgu Mulitang groups(3.25,6.5,13 g·kg-1), and fluoxetine group, with 20 rats in each group. Except normal group, the depression model was prepared through chronic unpredictable mild stimulation(CUMS). The normal group and the model group were given normal saline with 6.5 g·kg-1 by gavage. Chaihu Jia Longgu Mulitang groups were intragastrically given corresponding herbal drugs 3.75,6.5,13 g·kg-1, while fluoxetine group was intragastrically given fluoxetine 10 mg·kg-1 for 21 days, once a day. Then the depressive behaviors of rats were observed by sucrose preference test (SPT) and forced swimming test (FST). Western blot was used to detect the protein expressions of PI3K,Akt,GSK3β and phosphorylation level. Result:Compared with normal group,the sucrose preference index was decreased significantly,while the immobility time in FST was increased significantly(P<0.01), the protein expressions of PI3K, p-PI3K p110, p-PI3K p85 were decreased significantly (P<0.01), and expressions of Akt, p-Akt Thr308,p-Akt Ser473, p-GSK3β Ser9 and β-catenin were decreased significantly(P<0.01), while the level of GSK3β, p-GSK3β Tyr216 were increased significantly in model group(P<0.05,P<0.01). Compared with model group,Chaihu Jia Longgu Mulitang could increase sucrose preference index and decrease the immobility time in FST(P<0.01), the protein expressions of PI3K p110 and PI3K p85 was increased significantly (P<0.01), levels of Akt Thr308,Akt Ser473, p-GSK3β Ser9, β-catenin were increased significantly (P<0.01), whereas levels of GSK3β, and GSK3β Tyr216 were decreased significantly. Conclusion:Chaihu Jia Longgu Mulitang could increase protein expression and activity of PI3K in rat hippocampus, activate Akt, inhibit GSK3β kinase activity and prevent β-catenin from degradation, so as to increase PI3K/Akt pathway activity in rat hippocampus, and protect hippocampal neurons.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the ability of SD rat dental papillae cells forming dentin-like structure induced by millipore filter combined with transforming growth factor-β(1) (TGF-β(1)).</p><p><b>METHODS</b>The first passage SD rat dental papillae cells were enzymatically dissociated and centrifuged to obtain a cell mass. The cell mass was seeded on the millipore filter combined with TGF-β(1). The complex was incubated for 6 d in vitro or transplanted under the renal capsule for 2 weeks. Then the differentiation of dental papillae cells on the filter and the formation of mineral tissue on the implant were analyzed.</p><p><b>RESULTS</b>A layer of polarized columnar cells were observed along the surface of the millipore filter, with cell processes extending into the porous media. Dentin sialoprotein (DSP) and dentin matrix protein-1 (DMP-1) were positive in these cells. After 2 weeks, tubular dentin matrix was deposited on the surface of the aligned cells. Scanning electron microscopy showed that the thickness of newly formed tubular dentin was consistent. DSP and DMP-1 were expressed in columnar cells, tubular matrix and the dental papillae cells adjacent to the filter.</p><p><b>CONCLUSIONS</b>The millipore filter combined with TGF-β(1) could effectively recruit progenitors onto its surface and induce odontoblast differentiation, secrete matrix in a homogenous manner, leading to dentinogenesis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the primary result of finger flexion reconstruction in the procedure of direct anastomosis of contralateral C(7) transferred through the prespinal route with lower trunk in children suffered traumatic brachial plexus root avulsion injury.</p><p><b>METHODS</b>On the healthy side, the C(7) nerve root was identified anatomically and transected at the level of division by dissecting its anterior and posterior division as far distal as possible up to the level where the nerve fibers interweaving with other division, then the contralateral C(7) nerve root was dissected proximally up to the neuroforamina. The contralateral C(7) nerve root was transferred to contralateral side through the prespinal route. The entire brachial plexus of suffered side was exposed through the union incision superior and inferior to the clavicle, The lower trunk was identified and dissected proximally to the C(8) and T(1) nerve root which were severed at the lateral margin of anterior scalenus, and then the dorsal division and anterior medial pectoral nerve of lower trunk were severed. The median nerve, ulnar nerve and medial antebrachial cutaneous nerve were identified from the origin and dissected distally continue to the midpoint of upper arm, and lateral head of the median nerve was severed so that the lower trunk, medial cord and median nerve, ulnar nerve and medial antebrachial cutaneous nerve can be fully mobilized. Anteriorly flexion and adduction of the should at 0 degrees and flexion elbow at 90 degrees , this could allow considerable length to be gained when pulling the lower trunk proximally, direct anastomosis of contralateral C(7) with lower trunk was performed. If there was any tension exist, the appropriate humerus shorten osteotomy should be performed. From August 2004 to December 2008, 20 children including 13 cases with total brachial plexus nerve root avulsion injury and 7 cases with middle and lower trunk avulsion injury were repaired by this procedure. Twenty cases including 16 males and 4 females, the average age was 13 years with a range of 5 to 18 years. The interval from injury to operation ranged 1 to 11 months with a mean of 4.6 months. Eleven patients were performed the humeral shorten osteotomy, the length of the humeral shorten was 2.0 - 4.5 cm, with the mean of (3.1 +/- 0.7) cm.</p><p><b>RESULTS</b>The follow up period was 12 to 51 months, with the average of 26 months. The muscle strength of finger flexion attained M 4 in 18 cases, M 2 in 2 cases. The motor function of thumb flexion gained M 4 in 10 cases, M 3 in 8 cases, M2 in 2 cases. Of the 2 cases achieved motor function of intrinsic muscles of the hand of M3.</p><p><b>CONCLUSIONS</b>The direct anastomosis of contralateral C(7) with lower trunk in children with traumatic brachial plexus avulsion injury can improve the effect of reconstructing the function of finger flexion because it reduces one never anastomosis site and decreases the distance of nerve regeneration compared with the traditional method. With this modified procedure, the functional recovery of intrinsic muscles of the hand in children with traumatic brachial plexus avulsion injury is becoming possible.</p>

ABSTRACT

<p><b>OBJECTIVE</b>Studies have showed that L type calcium channel plays an important role in dentin calcification and affects tooth development and tooth reparation after injury. The objective of this article is to study the effects of nimodipine, blocking agent of L type calcium channel, on human dentinogenesis using human tooth slice organ culture in vitro.</p><p><b>METHODS</b>Young healthy human premolars were collected, and cut into 2 mm-thick transverse slices by low speed diamond saw. Agarose beads dipped in nimodipine solution and PBS weresy minetrically placed on tooth slices, and the slices were then embedded in a semisolid agarose-based medium and cultured with organ culture method for 1 week. Fluorescent band of tetracycline, Von-Kossa staining, immunohistochemical staining of the slices and transmission electron microscopy (TEM) of odontoblasts were observed to evaluate dentinogenesis changes of the slices.</p><p><b>RESULTS</b>Tooth slices were successfully cultured in vitro for 1 week and the odontoblasts could maintain their original morphology. After treatment with nimodipine, the fluorescent band of tetracycline was narrow and weak, and globular calcification in predentine was decreased compared with the control. TEM showed that secretory vesicles in odontoblast were somewhat increased, hut iminunohistochemical staining for collagen I showed no difference between the two groups.</p><p><b>CONCLUSION</b>Nimodipine can influence the calcification of dentine, but has no obvious influence on the synthesis and secretion of dentine matrix. The results show that L type calcium channel is important in dentin calcification.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the treatment of the displaced neck and subcapital fractures of the fifth metacarpal.</p><p><b>METHODS</b>Thirty-one patients with the neck and subcapital fractures of the fifth metacarpal had been operated on with antegrade intramedullary fixation between January and August 2005. There was 11 subcapital fractures (group A), and 20 neck fractures (group B). Following closed reduction of the fracture, a blunt 2.0 mm diameter K-wire, which was pre-bent into 20 degrees at the distal end, was inserted into the medullary canal of the fifth metacarpal and fixed the fractures. Postoperatively, patients in group A were immobilized in a short arm plaster splint for 4 weeks, and the ones in group B were treated with unrestricted mobilization.</p><p><b>RESULTS</b>Operative time was 18 min averagely (range 5 to 30 min). Twenty-nine of 31 patients obtained anatomic reduction, and 2 patients had 2/3 apposition of bone end and no rotational deformity. Follow-up was available for all patients. The average follow-up interval was 4 months, with a range of 3 - 6 months. The head/shaft angle of the fifth metacarpal in group A was 63.4 degrees +/- 14.5 degrees preoperatively, and 15.0 degrees +/- 2.5 degrees postoperatively, and 15.4 degrees +/- 2.6 degrees in 3 months postoperatively. The difference between preoperative and postoperative angles was highly significant. The range of motion of the metacarpal joint was 89.5 degrees +/- 4.3 degrees postoperatively, which was not significantly different compared with that of uninjured side. The head/shaft angle in group B was 59.1 degrees +/- 10.0 degrees preoperatively, and 15.9 degrees +/- 2.5 degrees postoperatively, and 15.5 degrees +/- 2.8 degrees in 3 months postoperatively. The difference between preoperative and postoperative angles was highly significant. The range of motion of the metacarpal joint was 88.6 degrees +/- 3.6 degrees postoperatively, which was not significantly different compared with that of uninjured side.</p><p><b>CONCLUSIONS</b>The technique is technically easy to perform, minimally invasive, low-cost, allowing early hand mobilization, with good functional results and low morbidity, and it has been proved to be an ideal method.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the prevalence of domestic violence (DV) in Hunan.</p><p><b>METHODS</b>Using a multi-stage sampling strategy, 9451 households involving 32 720 persons in urban, rural and industrial areas in Hunan, China were studied. Multiform clue investigation and face-to-face interviews were combined to investigate the prevalence of DV.</p><p><b>RESULTS</b>A lifetime prevalence of DV was reported by 1533 households (16.2%). A total of 1098 households (11.6%) reported at least one incident of DV in the previous year. Both lifetime and 12-month prevalence of DV varied significantly by geographic setting (P < 0.01). The lifetime prevalence abuse rates were: spousal 10.2%, child abuse 7.8%, and elder 1.5%. With regard to household structure, the lifetime prevalence of DV was highest among those remarried families (21.0%), followed by married couples with one child and extended families with several generations living together (20.1% and 20.0%, respectively). The highest rate of spousal abuse was found among remarried families (14.7%), while child and elder abuse was most prevalent among extended families (12.4% and 4.1%, respectively).</p><p><b>CONCLUSIONS</b>The findings suggested that although the prevalence of DV in Hunan was modest compared to Western countries, it remained a serious public health problem affecting over 1 in 10 households. Furthermore, the prevalence of various types of DV varied by geographic setting and family structure, suggesting that diverse geographic setting and family constellations carried different risk and protective features.</p>
